Introducing AECS-QuikPrep Ltd

First established in 1983 as AECS, we recently renamed to AECS-QuikPrep Ltd. We are after 25 years of trading, one of the longest established chromatography instrument design and production manufactures in the United Kingdom. Our Managing Director and co-founder, Dr Les Brown is still a practising chromatographer and has personally been involved in countercurrent instrumentation sales, design and the development of generic counter current / liquid-liquid chromatography method development strategies since the early 1990’s.

Mr Robin Self our Head of Engineering was previously a lead engineer at British Aerospace for Approximately 20 years. In the most recent eight years that Robin has controlled the Quattro CCC design and production, Robin has revolutionised function, fitness for purpose and reliability, plus brought modularity to the Quattro CCC product range.

Quattro CCC™ are fun! Prof Dra Gilda & Suzana Guimaraes Leitao, Joint Chairpersons of CCC2008, Rio, Brazil, celebrating the arrival of their Quattro HTPrep™ prior to installation in their laboratory. This HTPrep™ was used for the Seminar's Workshop on Liquid-Liquid Chromatography

The NEW HTPREP AND INTROPREP HT QUATTRO CCC Mk 6’s can both be used for rapid prep of small scale (sub mg to 100+ mg) and larger mass (100+ mg to multi g) resolution on a single Quattro CCC as they have at least two coils of different volumes and different id’s (typically a high resolution / low mass injection ~20 ml coil of 1.0 mm id and a higher mass injection ~100ml coil of 2.0 mm id). Both these instruments are also ideal for ccc-MS.

At CCC2008 the HTPrep purchased by the Federal University of Rio (shown above) was utlised to demonstrate CCC / CPC method principles to attendees of CCC2008 at the CCC / CPC practical workshop sessions. Our New HTPrep and IntroPrep HT were designed to meet the needs of  Major Pharmaceutical Company to exploit liquid-liquid chromatography in their High-Throughput Purification Group. We believe the first ever application of countercurrent chromatography for routine HT was given at CCC2008 by their staff after their novel HT research on their Quattro CCC HTPrep. This oral lecture will shortly be published in CCC2008 proceedings.

Shown above is an HTPrep with full automation capable of interaction with external computer systems, for integrated HT. This is the Quattro CCC presently used by the Major Pharmaceutical Company, which utilises their design of computer programme to control sample injection, HPLC pumping, Quattro CCC HTPrep, detector, fraction collection, and data system. Additional features to standard for this HTPrep included automation, solvent leak sensor, vibration sensor and bench lock-down.

Les & others will travel anywhere in the world to assist in CCC / CPC / HPLC research projects (see also our HPLC website www.chiral-hplc.com ). A few recent examples of training courses / co-research projects are Rio, Brasil 2001, Vicosa, Brasil 2004, Adelaide, Australia 2006, Chicago, USA, 2007, Guangzhou, China 2007 etc. We assisted in founding the bi-annual CCC200X series, which started in London in 2000, and the Rio-CCC2001 series. We were also the first commercial exhibitor sponsor for CCC2006 at NIH, USA, where we exhibited. We sponsored CCC2008 in Rio, Brasil and have been appointed a co-supporter for CCC2010 in Lyon. QUATTRO CCC INSTRUMENT TYPE DEFINITIONS

The new, low-cost IntroPrep series consists of the introductory Quattro CCC chassis option, and is offered with two bobbin pack options, one with a single coil, the other with a 2 coil options. Only PTFE coils are available on the IntroPrep series.

The QuikPrep™ high performance liquid-liquid chromatograph™ (HPL-LC™), introduces both PTFE and Stainless Steel coil options to countercurrent chromatography. QuikPrep offer enormous choice of coil options from 20 ml with 1 mm id coils up to ~750 + ml total capacity with large id’s. The former could be used for sub milligram preps and the later for preps up to 30 + g injected mass. Yet all this is possible within a minimally sized chassis (see pictures).  The QuikPrep has 2 bobbins per instrument and the option of either 2 or 4 coils per instrument. Coils of same id can be used in series or in parallel. All four coils could be doing different preps at the same time if required. Chose your chassis. Chose chassis options. Chose coil id and material. Chose number of coils. You design your Quattro CCC to fit your requirements. We build to your specifications (see brochures for options, link via thumbnails on left vertical bar).

The LabPrep approximately doubles the volume capacity options available on a QuikPrep. Shown below is a LabPrep CCC in a typical user system, inclusive of HPLC pump, injector and detector. Think of a Quattro CCC as a mechanical Prep HPLC column, and fit it into your HPLC system as you would a Prep HPLC column.

As an Quattro CCC often uses a third to a tenth of the solvent that might be used by a Flash or Prep HPLC for the same mass injected, standard analytical HPLC pumps, which pump up to 10 ml / min, may be utilised with a LabPrep being used for preparing multiple 10’s of grams of target containing matrix injected. This represents a massive solvent saving and has the convenience of utilising standard analytical HPLC pumps, which most labs have available. This saves the cost of buying dedicated Prep HPLC pumps, which would definitely be required even for 21 x 250 mm Prep HPLC columns. A 21 x 250 mm HPLC in reverse phase material with a typical flow requirement of 20 ml / min, would be expensive to purchase. A single contaminated sample might destroy it, plus it would only be capable of “on-column” loading of 100 to 1000 mg, rather than the 10’s g loading of a Quattro CCC utilising 10 ml / min flow rates. To achieve 1’s or 10’s grams injected, prep or process HPLC flows of 50 to 1000 ml / min might be required. Hardly any wonder we recommend the AECS designed Sequential CCC-HPLC generic gradient procedures. Inject crude extract on to a Quattro CCC, run a generic gradient. Each CCC fraction would have such a small polarity range that an isocratic HPLC run would be suitable, thereby massively reducing the risk of HPLC column contamination. Plus the target to contaminant mass in the Quattro CCC derived fraction would probably reflect a 50 to 95+ % target purity already by Quattro CCC. If insufficient purity was achieved by Quattro CCC alone, the relatively low mass / high purity of the CCC fraction would then make, sequential, final prep HPLC very cost-effective, with low risk of HPLC column contamination. We are one of the longest established liquid-liquid chromatograph specialists in the world. Recessions have come, and recessions have gone in last 25 years, AECS is still here and will still be here for you a long time in the future. We have always been self-financing. We have existed for over 25 years based on commercial and profitable trading. Certain of our much more recent CCC competitors exist mainly by Venture Capital & Bank Loan cash injection support and collaborative Grants with an associated University, rather than by profit income. Neither Venture Capital nor Grants are stable or long-term, especially in the present World Economic climate. From the early 1990’s to 2000 we utilised standard instrument design techniques for our Quattro CCC. Feedback from customers showed us this was inappropriate in liquid-liquid chromatography, so we assigned this technology to Brunel University, UK, subject to a fee being paid and certain conditions. We collaborated with BIB / Brunel University in CCC research projects from 1993 to 2000, but then chose to cease all future collaboration as they became competitors in 2000, then more recently, shareholders in a start-up UK Ltd that Brunel University and it’s Professor/staff co-founded. Other collaborations have included, AstraZeneca, GSK, Shell Research, Syngenta (then AZ Agrochemicals), University College of Swansea and University College of London.

Based on our own internal research we developed “in-house” a new MODULAR CONCEPT for the Quattro CCC Mk IV, which was released in 2002. From 2002 all Quattro CCC now offer our unique modularity. All Quattro CCC from the 20 ml ccc-MSPrep coil to a 100+ litres ProcessPrep share the same planet & sun radii, beta value ranges, speed range options etc. This design concept was further updated to the Quattro Mk 5 design in 2005.

Unlike conventional high pressure liquid chromatography (HPLC), low pressure liquid chromatography and flash chromatography, which are all solid-liquid chromatography systems, SLC ……… With countercurrent chromatography, the chromatographic process utilises one immiscible, immobilised, stationary liquid, and another immiscible, flowing mobile liquid, thus a more accurate name than counter current chromatography would be, high performance liquid-liquid chromatography, HPL-LC™.

To summarise, one of the pair of immiscible, biphasic liquids is immobilised in a planetary centrifuge of a Quattro CCC™ design, and the other flows over this, periodically mixing with and separating from the stationary liquid

Imagine taking a separating funnel and doing 100,000 separations in an hour……….a Quattro CCC™ does the equivalent of this.

Counter-current chromatography can separate diasteromers, chiral & non-chiral API’s, natural products, synthesized compounds, petrochemical, inorganic anions and cations, precious metals and radio active compounds without the risk of “on-column degradation" or "infinite adsorption".


The Quattro CCC™ range has been designed as a modular system, which allows predictable scale up from sub-milligram on a minimum footprint laboratory instrument, to a multi-rotor simulated moving bed countercurrent chromatography multiple tonne per annum system, designed to work in a process, intrinsically safe environment.  
      
The major benefits of counter-current chromatography are : “No Infinite Adsorption”; “No On Column Degradation", ”No Need For Expensive Stationary Chromatographic Phase”, “Can Be Inorganic and/or Organic; The Inorganic can be Metal, Non Metal or Radionucleotides; The Organic can be, Chiral or non-Chiral, Reverse or Normal Phase, Miscelle or Reverse Miscelle, Liquid Ion Exchange", “From Infinity Polar or Infinity Non Polar”, “One Quattro CCC Could Be Performing Up To 4 Separate Preparations”, “Can Prepare From Crude Extract Narrow Polarity Ranges Suitable For Prep HPLC Without Fear Of Contaminating The HPLC Column” and often a “Reduction Of 3 To 10 Times In The Solvent Usage, As Compared To Preparative Flash Or HPLC”.

Traditional, sub-critical (HFC’s) and super critical fluid (CO2) solvents may all be utilized in chiral & non-chiral counter current chromatography.
