From the above picture it can be seen that the NEW
Quattro CCC™ Mk 5 LabPrep™ is basically a simple addition
to any standard solid / liquid chromatography instrumentation set-up.
It makes no difference if you are adding CCC on laboratory or process
scale, or to either a high or low pressure chromatography system, you
simply treat the CCC as a column. You utilise your standard non-column
ancillary components, ie injector ( near CCC ), pump, switching valves,
detector, fraction collector, control and data system etc as you would
normally
The only difference between utilising a prep HPLC or Flash column and
a CCC is that in CCC the column is a mechanical instrument, with a tube
filled with your chosen two immiscible liquids, instead of a tube filled
with your selection of solid particles and a liquid.
BUT CCC has the HUGE ADVANTAGE that it cannot be poisoned
like an expensive HPLC ( or cheaper Flash ) columns. Plus you can have
a new column for each preparation for only the cost of the solvent.
In addition, a typical Quattro CCC™ preparation will use much
lower flow rates and considerably less solvent than either Prep HPLC
or Flash for the same mass injected.
As you can see from above, a Quattro CCC™ Mk
5 LabPrep™ instrument is not much bigger than a standard lab HPLC
pump etc, yet is capable of 5 to 20g ( occasionally up to 40 g ) per
1000 ml capacity, but with laboratory scale of flow rates, that are
readily available from a simple, standard analytical, 10 ml / min laboratory
type HPLC system.
As you know for high resolution, 5 to 40 g loadings,
it would require large flow HPLC prep pumps ( 100 to 1000+ mls per min
), an enormous amount more solvent, and expensive prep process scale
hplc columns, with expensive axial or radial compression process column
hardware system. At this 5 to 40 g loading scale for high resolution
prep HPLC prep, usually your equipment requires expensive hardware to
intrinsically safe standard, because of the large volume of solvents
used. It is a EEC requirement by law to utilise Intrinsically Safe Instrumentation
if total organic solvent usage exceeds 5000ml for the process. This
is a considerable restriction for Flash and Prep HPLC, but much less
so in CCC.
For
those not familiar with Prep HPLC column loading capacity, we have included
a comparison of "On-Column Loadings" for Baseline Resolution
/ Simple Heart-Cut / Prep Overload + Typical Flow Rates for a variety
of laboratory scale HPLC prep columns ( bore in mm x length in mm ).

As you can see for pre-packed prep HPLC columns, the
largest of which is typically 50 mm id, the "Overload Injection
Loading" is only just over 2 grams, and yet you could already be
utilising flows of 100 ml / min. Such a prep could probably be achieved
with a 100 to 400 ml Quattro CCC™ coil, with flows of only 2 to
10 ml / min, and a very considerably reduction in total solvent usage

Scale this up to Process Scale Chromatography,
and the cost savings are very
considerable, for both ancillary equipment and solvent usage.
With
CCC, equivalent prep resolution is available MUCH MORE CHEAPLY, and
one CCC can be reverse or normal or ion exchange, or ligand / affinity
etc at fraction purchase or running cost, plus you can renew column
contents each injection, and easily wash / sterilise between new preparations
to meet GMP.

What is a PrepPlus™ ?
A PrepPlus™ is a additional, cased rotor module, that simply bolts
to the side of your existing Quattro CCC™ Mk 5 and is controlled
and driven by your existing motor and control electronics. This allows
low-cost, very cost-effective, modular expansion of the capacity and throughput
of all Quattro CCC™ Mk 5's; a feature unique to the Quattro CCC™
Mk 5
For both the QuikPrep™ and LabPrep™, their PrepPlus™
are aluminium constructed, LabPrep™ sized modules, thus the capacity
could be increased by up to 1600 ml, to give a total capacity of a LabPrep™
+ PrepPlus™ of approximately 3200ml with 16 to 64 g loading capacity.
For the PilotPrep™, this PrepPlus™ is a stainless steel constructed
PilotPrep™ sized module, thus the capacity could be increased by
up to 3200 ml, to give a total capacity of a PilotPrep™ + PrepPlus™
of approximately 6400ml with 32 to 128 g loading capacity. As the flow
rates used in a Quattro CCC™ are a fraction of that used in a process
HPLC, the overall cost of 100g capacity CCC is also much less expensive
that a 1000ml/min prep HPLC installation capable of 100+g "on-column"
loadings, inclusive of pump, hplc column, etc, PLUS YOU HAVE NO
ONGOING SOLID HPLC STATIONARY PHASE COSTS, OR CHANCE TO POISON YOUR SOLID
STATIONARY PHASE, OR FOR THIS SOLID PHASE TO ADSORB / CHEMICALLY ALTER
